Variations between individuals and inter-regions in the activity of enteric drug metabolism Activities: Results with the Human Intestinal Mucosa epithelium CryoPreserved (Chim) from the small intestine of 14 donors

We have previously reported successful insulation and cryopreservation of human intestinal mucosa (CHIM) with the viability retention and activity of drug metabolism enzymes. Here we report the results of quantification of drug metabolism activities in Chim from various regions in the small intestine of 14 individual donors. Chim isolated from the duodenum, jejunum, and ileum of 10 individuals, and from 10 consecutive 12-inch segments ranging from pyloric sfingter from the small human intestine of four additional individuals.
P450 and non-P450 medicine metabolization enzymes (CYP1A2, CYP2C8, CYP2B6, CYP2C8, CYP2C9, CYP2C19, CYP2D6, CYP2E1, CYP3A, UGT, Sust, AO, NAT1, and NAT2) are measured through selective substrate incubation. Activities that can be measured are observed for all lanes except for CYP2A6. Comparison of duodenal, jejunum, and ileum in 10 donors showing Jejunum has a higher activity for CYP2C9, CYP3A, UGT, SULT, MAO, and NAT1. More definitions of regional variations with Chim from ten proximal small intestinal segments show that the segment immediately after the first 12-inch segment (duodenum) has the highest activity for most drug metabolism enzymes but with a substantial difference in four donors. Our overall results show that there are substantial individual differences in drug metabolism enzymes and that Jejunum, especially the region as soon as the duodenum, has the highest drug metabolic enzyme activity.
The effects of leukocyte anti-human antigen immunity sensitization from allographic cryopreserved and blood transfusions
Allotransplansis of vascular composite (VCA) is the most sophisticated reconstructive technique available for patients who suffer devastating burns into legs or faces. However, VCA requires matching donors. Burn patients have reportedly experienced sensitization, or the development of anti-human antigen antigens leukocyte, during resuscitation and wound coverage, potentially blocking them from future VCA. This study strives to investigate blood and allographical contributions to sensitization in burn patients. Four groups compared: burns of burns that accept blood products and allograft (group 1), burn patients who only accept blood products (group 2), trauma patients only accept blood products (group 3), and healthy volunteer control (Group 4). The average calculation of antibodies of the reactive panel (shows sensitization) is higher in group 1 compared to group 4 (p = 0.035).
In addition, the incidence of severe severe sensitization in group 1 was relative to other groups (p = 0.049). When comparing groups of patients who do not have sensitization, minor sensitization, moderate sensitization, and severe sensitization, there is no significant difference in age, gender, blood products received, the total body surface area burned, or allograft used between groups, though Very sensitive patients tend to have the involvement of a larger body surface area and receive more packaged red blood cell units and allograft (p = 0.079, p = 0.196, and p = 0.072, respectively). Therefore we conclude that while burning patients who receive allograft and blood show a higher incidence of the sensitization of leukocyte anti-human antigens relative to healthy control, this difference cannot be only associated with the use of allograft or transfusions.
Application of Cryopreserved Human Intestinal Mucosa and Human Enterocyt CryoPreserved in the Evaluation of Herb-Drug Interaction: Evaluation of the Potential inhibition of CYP3A Juice Balinese Juice and Twenty-Nine Commercial Formulation Herbal Supplements
Commercial Formulations 29 Herbal supplements commonly used (HS) and grapefruit juice are evaluated for potential drug interactions through the quantification of their CYP3A inhibition potential in two experimental models in vitro from the small intestine of humans, cryopreserved human intestinal mucosa and the Human Enterocyte CryoPreserved (CHE). Two CYP3A substrates are used – in studies with Chim, CYP3A activity is quantified through the quantification of LC / MS MSZolam 1′-hydroxylation while in CHE, Luciferin Science metabolism to Luciferin is quantified by luminescence.
After Chim’s treatment with the estimated concentration of HS lumen in every oral administration (the suggested dose was dissolved in 200 ml of culture media),> 80% inhibition of CYP3A was observed for green tea extract, Valeria roots, St. John, horehound juice and grapefruit. Obstacles of less than 50% observed for fenugreek, aloe vera, guarana, soybean isoflavone, maca, echinacea, spirulina, evening primrose, milk thistle, cranberry, ria yeast, rhodiola, ginkgo biloba, turmeric, curcumin, white beans, garlic, garlic, Cinnamon, see palmetto berry fruit, panax ginseng, black grass juice, wheat grass juice, flaxseed oil, black cohosh, and ginger root. The results are confirmed in the study of dose-responses with HS obtained from three suppliers for four HS inhibition (green tea extract, horehound, St. John’s Wort, Root Valerian), and three HS Black Elderberry (Black Cohosh, Black Elderberry), Echinacea).
Similar results were obtained with HS inhibitors in Che. The results illustrate that Chim and Che represent an experimental model in vitro which is physiologically relevant to evaluate the potential for herbal supplements drug interactions. Significance Statement in Evaluation in Vitro 29 Popular Herbal Supplements in Cryopreserved Human Intestinal Mucosa Identifying Green Tea Extract, Horehound, St. John’s Wort, Root Valerian to have the potential for inhibition of CYP3A similar to grapefruit juice, indicates their potential to have a clinically significant pharmacokinetics. Interaction with orally managed drugs is the CYP3A substrate. The results showed that Chim could be used for evaluating in vitro drug interactions involving enteric drug metabolism.
Extract Vulgare Origanum Aqueous improves the quality of human spermatozoa cryopreserved through its antioxidant effects
Excessive production of reactive oxygen species (ROS) during cement cryopreservation can cause structural and functional changes in spermatozoa. It is known that antioxidants can reduce the ROS effect. In addition, the application of antioxidants in media freezing is the right strategy to protect spermatozoa to the adverse effects of ROS during the cryopreservation process. For example, Oregano is a medicinal plant with important activities, with antiseptic properties, antibacterial, antitrombotic, and antioxidants.

This study aims to evaluate the antioxidant effects of oregano extract in the human spermatozoa cryopreserved. In the first phase, 13 cement samples with different oregano extract concentrations (0.0, 50, 100, 150, 300, and 500 μg / mL) were cryopreserved to achieve the optimal dose of oregano extract. Then, the motility, feasibility, and integrity of plasma membranes is evaluated.
