The level of disbursement of iliaca artery of the human kriopreserved allografts (CHIAA) directly affects the fatigue of structural changes that occur during this process.
Experiments were carried out at ten Chiaa. 10% dimethylsulphoxide in hydroxyethyl 6% starch solution is used as cryoprotectant; All Chiaa is cooled at a level controlled and stored in the liquid nitrogen vapor phase (-194 ° C). Two Disbursement Protocols are tested: (1) placing Chiaa in a water bath at 37 ° C, and (2) Chiaa is disbursed in a controlled environment at 5 ° C. All samples undergo analysis under the scanning electron microscope. Testing of Chiaa’s mechanical properties is evaluated on a single-made strain test engine specifically made. Longitudinal and circular samples were prepared from each Chiaa tested.
Results: Ultrastructoral analysis revealed that the five Chiaa was disbursed during the Disbursement Protocol 1 which showed more damage to the subendotial structure when compared to the disbursed sample in the protocol 2. Mechanical properties: 1-Longitudinal Disbursement Protocol UTS 2, 53 ± 0, 47 MPa on relative strain 1, 27 ± 0, 12 and around UTS 1, 94 ± 0, 27 MPa on the relative strain 1, 33 ± 0, 09. Disbursement Protocol 2-Longitudinal Tensile Strain (UTS) 2, 42 ± 0, 34 MPa on relative strain 1, 32 ± 0, 09 and circular utiat 1, 98 ± 0, 26 MPa on the relative strain of 1, 29 ± 0, 07. Comparing UTS does not show statistical differences between disbursement methods.
Expand the use of test challenges as Biomarkers repair DNA in the human population: protocols for fresh blood samples and cryopreserved, and for different challenges agents
Deficiencies in DNA (DDRR) damage response (DDRR) can cause serious pathological results; Therefore, having the ability to determine individual DDRRs will increase concerns in the assessment of health risks and in determining individual responses to cancer therapy. However, most methods for evaluating DDRR are not fully suitable for population studies. Test challenges have received acceptance for this purpose. The test traditionally uses X-rays as a challenge agent and mononuclear cell peripheral blood isolated (PBMC) as cell specimens. To increase the use of the test, the purpose of this investigation is to use processed blood samples differently, to hire other challenges agents with various induction mechanisms of DNA damage / improvement, and to produce protocols to detect different DDRR capacities.
Fresh and frozen blood samples are challenged by bleomycin, methyl methanesulfonate (MMS) and ultraviolet light. Induction significant damage after all treatments, and progressive DDRR and depends on time observed. There was no significant difference obtained in the capacity of DDRR of blood samples all fresh or frozen compared to PBMC, except that fresh blood samples showed the capacity of DDRR induced by MMS higher than PBMC. The results of this study indicate that the test of comets can be used as routine biomarkers of DDRR capacity in human biomonitor studies, and that all blood is also a biomatrix that is useful for this test. Data collected allows us to recommend various protocols for test-comets that are useful for evaluating DDRR capacity in several main DNA repair lines. As a result, the usefulness of testing the challenges can be greatly expanded.
Micronucleus cytokinesis-block test on fresh blood mononuclear cells and isolated human cryopreserved
The Cytokinesis-Block Micronucleus (CBMN) test is a standard method used for genotoxicity studies. Conventional overall blood culture (WBC) is often used for this test, although this test can also be carried out on isolated blood mononuclear cell culture (PBMC). However, the standardization of the protocol for the CBMN PBMC test has not been widely investigated. The purpose of this study was to optimize the reliable CBMN test protocol for fresh peripheral blood mononuclear cells and cryopreserved (PBMC), and to compare the results of Micronuclei (MNI) between WBC and PBMC culture.
The CBMN G0 test is carried out in all blood, new isolated, and PBMC cryopreserved from healthy human blood samples and five samples of radiosensitive patients. Cells hit by 220 kv x-ray in vitro ranging from 0.5 to 2 gy. The Optimized PBMC CBMN test shows adequate repetition and small inter-individual variability. MNI value is significantly higher for WBC than for fresh PBMC. In addition, the Cryopreservation of PBMC resulted in an increase in significant MNI values, while different Cryopreservation time did not have a significant impact. In conclusion, our standard CBMN test on fresh PBMC and cryopreserved can be used for genotoxicity, biological dosimmetry, and radiosensitivity assessment.
Bilaminar Engineering with Corriocer Advanced Flap and Cryopreserved Human Amniotic Membranes in the treatment of gingiva recession
Gingiva recession is usually treated with surgical therapy that involves the use of connective network autograft to thicken gingiva network. It has an important biological impact on patients because they will have surgical wounds on two different oral cavity sites. In this case report, we offer an alternative technique for the treatment of Gingiva recession class Miller. A 40-year-old woman patient has been treated with a combination of Bilaminar and Flap Flap technique in a coron to cover the Miller I Gingiva recession with the addition of the cryopreserved human amniotic membrane.

Description: Tonsil epithelium is a heterogeneous lymphoid organ containing two different types of actively differentiating epithelia, the lining stratified squamous epithelium and the reticulated crypt, or lymphoepithelium. The tonsil crypts represent a specialized compartment, important in the immunological functions of the tonsil because of the immediate proximity of the epithelial and lymphoid tissues. In Situ staining of tonsil epithelium suggests that expression of the simple epithelial keratins K8, K18 and K19 is located to the crypt epithelial cells. Cell culture of tonsil epithelium produces heterogeneous squamous epithelial culture in which differentiation appears to be ongoing and keratin expression patterns are consistent with those found in tonsil epithelial cells in situ. Description: Thymic epithelial cells (TyEpiC) are vital regulators of thymocyte development and T lymphocyte (T-cell) tolerance. The thymus contains two types of TyEpiC, cortical and medullary thymic epithelial cells, which regulate the positive selection of thymocytes and the negative selection of autoreactive T-cells. The inability of TyEpiC to remove autoreactive T-cells in the thymus can affect self-tolerance and contribute to the development of autoimmune diseases such as myasthenia gravis, type 1 diabetes, rheumatoid arthritis, and multiple sclerosis [3]. Studies have shown that changes in the expression of tumor necrosis factor receptor family members and transcription factors Foxn1 and autoimmune regulator (Aire) can alter the thymic microenvironment and self-tolerance. Description: The cornea is a unique tissue for two reasons: the transparency function and the synthesis of most of its proteins due to the cornea being an avascular tissue. The cornea consists of three distinct cell layers, the outer epithelium, the inner endothelium and the central stroma (the keratocytes). The corneal epithelium plays a role in the innate immune response by sensing the presence of pathogens and providing signals that activate the corneal defense system.
