Objective: To explore the role of miR-1193 / Janus kinase 3 (JAK3) axis and the potential mechanisms in rheumatoid arthritis (RA). Methods: The dysregulated genes and microRNAs (miRNAs) are filtered using a dataset of GSE12021 and GSE72564, whereas miRNA upstream of JAK3 was estimated by TargetScan. MH7A Then the cells were treated with interleukin-1β (IL-1β) to induce localized inflammation.
Dual luciferase report assay was used to estimate the relationship between JAK3 and miR-1193. flow cytometry and 3- [4,5-dimethylthiazol-2-yl] -2,5 diphenyl tetrazolium bromide (MTT) tests taken to analyze apoptosis and cell proliferation MH7A with inflammation-induced IL-1β, respectively. The relative expression of genes and proteins were detected by real-time quantitative polymerase chain reaction and western blot analysis. Finally, rescue experiments were employed to explore the effects of miR-1193 / JAK3 axis in inflammation-induced IL-1β.
Results: JAK3 was mainly up-regulated in RA, and targeted and negatively regulated by miR-1193 were lower expressed in RA tissue and cell IL-1β-treated. MiR-1193 significantly inhibited whilst mimicking mir 1193 MH7A inhibitor of cell proliferation promoted by inflammation-induced IL-1β. Furthermore, overexpression of JAK3 served auxo-action while depletion of JAK3 exhibited an inhibitory effect on cell proliferation MH7A with inflammation-induced IL-1β, which can be attenuated by miR-1193 and miR-1193 imitate inhibitor, respectively. Analog, JAK3 recovered cell proliferation is inhibited by miR-1193 imitate and inhibits cell apoptosis enhanced by mimicking mir 1193.
Conclusion: Up-regulation of miR-1193 suppressed proliferation and accelerated apoptosis MH7A cells with IL-1β inflammation-induced through targeting JAK3, which may provide a new understanding of the mechanisms underlying RA.

Rheumatoid arthritis (RA) is a chronic systemic autoimmune disease. New evidence shows that linc02381 suppressed development of colorectal cancer by regulating PI3 K signaling pathway, but linc02381 role in other diseases, such as RA, is still unclear. This study aims to reveal the mechanism of linc02381 in the development of RA. In vivo and in vitro, we found that linc02381 is upregulated in RA synovial tissue or RA fibroblast-like synoviocytes (RA-FLSs, P <0.01), which was detected by real-time quantitative polymerase chain reaction.
Cells Counting Kit-8, EDU, and Transwell tests reveal that the enhanced linc02381 excessive cell proliferation and invasion, and linc02381 knockdown inhibits cell proliferation and invasion in FLSs. In addition, the results of bioinformatics analysis, the luciferase reporter gene assay, and pull-down assay that can be directly verified linc02381 binds with miR-590-5p. MiR-590-5p was downregulated in RA-FLSs, and overexpression of linc02381 suppressed the expression of miR-590-5p that record-transcriptionally repressed expression of mitogen-activated protein kinase kinase 3 (MAP2K3), and overexpression of miR-590- 5p reversed excessive linc02381 effect on MAP2K3 expression.

MiR-590-5p inhibitor reversed the inhibitory effect of linc02381 knockdown on the proliferation and invasion FLSs, which increases the expression of MAP2K3, and activation of p38 and AP-1 in the MAPK signaling pathway. In short, linc02381 is upregulated in RA synovial tissue and RA-FLSs and RA exacerbated by absorbing miR-590-5p to activate the MAPK signaling pathway.
